Pthumeru Ihyll Chalice is Chalice Item in Bloodborne. Combining the Materials listed below in the Chalice will create a Chalice Dungeon. You must be at the Tomb Altar in Hunter's Dream to do this. This dungeon never changes Bosses, design or loot. Yharnam, Pthumerian Queen is located at the end of this Chalice Dungeon .

- In the pre-area side-dungeon of layer 3, at the end of the first hallway will be a bath messenger from which the Sinister Pthumeru Ihyll Root Chalice can be bought for 16,000 blood echoes. Two Kin Coldblood (12) are immediately nearby.

Today I join the ranks of those who have defeated Queen Yharnam. Just barely beat her first try, with zero blood vials left. I had also beaten the headless Bloodletting Beast first try. I actually had the most trouble with Pthumerian Descendant. He took either three or four tries. Still have to do the Isz dungeon, but it definitely seems that the defiled Pthumeru dungeon was the worst fixed one so far.

for anyone wondering/worrying after seeing that comment thread about grinding for ritual materials:
to put it simply, you don't need to grind for any items to finish the story chalices and get the platinum trophy. you just have to go through ng+, which you are meant to do anyway. you are intended to beat the depth 1 pthumerian chalice basically as soon as you get it (although you don't have to by any means; if you play it later it will just be really easy), but the cursed chalices and higher depth chalices are meant to be played when you are over level 100. if you play at this pace and just do the story chalices (pthumeru, central pthumeru, ailing loran, lower pthumeru, defiled chalice, and pthumeru ihyll chalice) you won't need to grind for anything. in any case, you need to get all three endings to get all three of the ending trophies.
think of it this way: if you are going to get the platinum trophy, you should have mastered the game. if you play a ng+ and you really know where all of the items are and what they're good for, then it's not a matter of grinding, it's a matter of beating that ng+ and actually mastering the game. i think if you are into the combat and weapon/stat system, then it will at least be fun to beat the game multiple times and try different strats on the bosses. if you are looking for the plat trophy, this is probably the kind of player you are already. if you are going to be grinding, it will only be for vials/levels when you need them for a boss.
personally, i did two new games: a run where i just did whatever and tried out the arcane tools and various weapons; then another where i made a specific build and rushed a specific weapon to beat the game with. on this latter file, i finished the hidden ending, just started new game+, got the weapon trophy, and intend to finish a 2nd ending and the queen eventually. i also finished the dlc and got all those trophies with this file. then i'll go back and get the 3rd ending by finishing that initial new game i started with the scuffed character build (i'm already on the wet nurse anyway).

Ughh...I was so close to being done with these godforsaken chalices for the Platinum. One of the worst experiences in all of Soulsborne. But no, I need 2 more Red Jellies to get in, which means having to do a ritual for Lower Loran...which means using up the materials I need for Great Pthumeru..which means not only do I have to do another f*cking chalice just for the Jelly, but I'll then have to farm MORE material to get into Great Pthumeru. God Dammit. These stupid chalices just will never end. There was a light at the end of the tunnel, just for a steel door to be slammed down in front of me. It's a cool idea in concept, but because of the lazy, repetitive design, mind numbingly frustrating, unfair Defiled Chalice and large amount of materials you need to get in, it's just the most drawn out, tedious thing in the series. It makes getting the Platinum for my favorite game ever made just an absolute nightmare.

Three of us stright up *cough* raped her in her room
2 or 3 heavy attack can stun lock her, and three of us just keeps stunning and stunning her

3rd layer lever is the easiest if you call for the old hunter: Olek bell is on the left of the room you first enter from the lantern, when you go down there're 2 pickaxe guys one can be visceral, then you will enter the big room kill the scorpion fast don't let the Kirkhammer guy notice you( call Wallar to make it easier if you haven't called Olek, his bell is behind the enemy hunter) then kill him and there will be some more scorpions and 2 flaming greatsword guys ahead, climbing up the ladder run through the bridge (there's a guy here careful not to fall down)and you will enter a room which may seem empty but there're 2 gargoyles here one is kind of in front of you and the other is upstairs with the gun merciless watcher and the lever. If you kill all of the mobs I mention you will net at least 400k blood with no moon runes. Oh and there's a bloodsucker when I go back to the ladder in the first room and I had no idea where did he come from.

When you go down the 3rd layer and before you reach the lantern, open the door on the left climb up enter the room climb up the ladder again there will be 2 kin coldblood(12) on the right with a mob in front the door on the left, after that you will enter a balcony open area, run around and eventually you will see a great one coldblood which gives you 40k blood.

2nd layer lever is pretty straightforward: go upstairs, enter the balcony open area, jump down, use the blue elixir and run to the staircase, which use to go up to the middle big platform and has dogs and mobs standing on, there's a room on the left with a mace merciless watcher and a lot of mobs, use another blue elixir if needed and rush in turn the lever then run the hell away from there and use bold hunter mark or die to them if you have little blood echoes.

The lever on 1st layer can be reached by entering the room with the merciless wathcer and dogs( use blue elixir and blood cocktail to avoid combat) open the door on the left inside the room there's a ghost kill it then climb up and the open door there are 2 rooms here one is empty the other has a sword merciless watcher and the lever. To cheese the first boss is super easy, use burial blade scythe form and charge attack him behind a pillar, he’ll never touch you and you can deal a decent amount of attack, just remember lure him to the front of the pillar and you get behind of it.

Anyone want to help me out I’m BL 150 I’ve gotten past the Pthumerian descendant but can’t get pass the headless bloodletting beast I’ve been close 3 times with just slivers of his hp left but end up cracking. Im just so tired of these chalice dungeons and this boss in general. I’ve tried ringing the beckoning bell for the past couple of days and I haven’t had any luck on getting an answer on the other end. My id is torresh123.

I just beat the first layer boss in my first try. If u can parry him 4 or 5 times , he can die easly. Use pillars to hide and w8 a good parry moment. And ofc he can throw his blade , when u see that attack just jump b and it cant hit u. Hope so this tips can be helpful.

Is this place harder than Lower Loran? That one's depth 5 and i'm able to handle that on my own at BL 80. Like, relatively easily. Though i have some good gems on my Threaded Cane and i know how to make a build as i've played a lot of dark souls. I just needed some help on the bosses, though i'm pretty sure i could've killed them solo with patience. Hopefully this place drops radial gems, Lower Loran only dropped waxing gems... Got a cool bolt abyssal waxing gem though.

The online for these chalice dungeons sucks. I can't connect to anybody needing help, nor can I connect with someone to help me.
I've played through majority of them solo. What happened to old fashioned 'summon signs'? This game was made by the makers of Demons Souls which invented that summon system. Whoever came up with the bells should be banned from future projects.

Contrary to some comments not only do you not need to farm materials, you do not need to go to ng+ to get all the materials either. Everything can be found as treasure either in ng or in the chalice dungeons themselves. You don’t need to do any dungeon twice either. You only have to explore thoroughly. Don’t forget the bonus side areas!
I didn’t even need to go to Isz.
